Direct Sequencing from Intact Bacterial Genomes
We have had some success with sequencing directly from bacterial genomes (i.e.
without an initial PCR reaction to amplify specific template regions). The
procedure is essentially the same as the one used to sequence from BAC DNA (please see
Sequencing from Large-Insert Clones).
For purification of bacterial genomic DNA, I have been told that Qiagen makes
a product for this purpose, and that the resulting DNA works well for direct
genomic sequencing.
